Chelsea got the better of Real over two legs in the semi-finals of last season's competition, while City will take on Atletico for the very first time in European competition.
Liverpool - who having won the Carabao Cup last month are still on course for an unprecedented quadruple - will be the happiest of the English sides with the draw, although they did lose to Benfica the last time the clubs met in the Champions League in 2006.
After Manchester United crashed out to Atletico Madrid on Tuesday, Chelsea became the third and final one of the Premier League trio through to the last eight when they saw off French side Lille on Wednesday.
The Blues, who are the Champions League holders, advanced 4-1 on aggregate despite the ongoing uncertainty around the club in the wake of the sanctions against owner Roman Abramovich.
Manager Thomas Tuchel praised the spirit of his players following the win.
"Chelsea has this kind of culture within the club, the building we work every day. What I found from day one is football first mentality," he said.

"Everybody pushes the first team to the limit, focused on football and football first. Then we try to implement the atmosphere. This culture is already installed and helps us to focus in difficult, distracting times.
"We need to encourage the players of course but I am also just a part, it feels so good we can produce results. I am very proud."
Manchester City, who were the beaten finalists in last year's competition, strolled into the last eight with a 5-0 aggregate win over Sporting Lisbon, with all of their goals coming in the first leg in the Portuguese capital.
Six times winners Liverpool also did the bulk of their work in the first leg, winning 2-0 at Inter Milan before 1-0 defeat to the Italian champions at Anfield.
